10号染色体缺失的磷酸酶及张力蛋白同源物((Phosphatase and tensin homolog deleted on chromosome ten, PTEN)为1997年新发现的抑癌基因，位于10q23.3，其转录产物是一种膜结合型脂质磷酸酶，作为肿瘤抑制因子在多种系统中下调PIP3的表达，在胚胎发育、细胞生长、分化、凋亡和迁移的调控中发挥重要的作用。PTEN具有PI3K活性，能拮抗PI3K的作用，通过催化PIP3的3位脱磷酸下调PIP3的水平。
